Diazepam is a benzodiazepine that enhances the effect of the inhibitory neurotransmitter GABA at GABA-A receptors. This reduces excessive nerve activity in the brain, producing anxiolytic, sedative, muscle-relaxant and anticonvulsant effects.
Tablet for oral use. Take exactly as prescribed by the doctor, usually 1–3 times daily depending on condition; with or without food. Swallow with water; avoid driving/operating machinery. Do not stop suddenly after regular use—taper as advised.
Drowsiness, dizziness, fatigue, muscle weakness, impaired coordination/ataxia, confusion, memory problems, slurred speech, nausea
Prescription-only; may cause dependence, tolerance and withdrawal—use the lowest effective dose for the shortest duration. Can cause marked sedation and respiratory depression, especially with opioids, alcohol or other CNS depressants; risk of falls (elderly). Use with caution in liver disease, respiratory disease (COPD/sleep apnoea), depression/suicidal risk, history of substance abuse, myasthenia gravis, and in elderly/debilitated patients. Do not abruptly discontinue after prolonged use. Not recommended in severe hepatic impairment; caution in glaucoma (narrow-angle).
Alcohol, opioids (e.g., morphine, tramadol), other sedatives/hypnotics, antipsychotics, antidepressants, antihistamines (sedating) (additive CNS depression); CYP3A4/CYP2C19 inhibitors (e.g., ketoconazole, itraconazole, fluconazole, erythromycin/clarithromycin, cimetidine, omeprazole) may increase diazepam levels; CYP inducers (e.g., rifampicin, carbamazepine, phenytoin, phenobarbital) may reduce effect; valproate may increase sedation.
