Overview
Drug | Injection
Uses & Benefits
Short-term management of severe anxiety and agitation, acute seizures/status epilepticus, premedication and procedural sedation, muscle spasm/spasticity (e.g., tetanus), alcohol withdrawal symptoms (including delirium tremens)
How It Works
Diazepam is a benzodiazepine that enhances the effect of GABA (an inhibitory neurotransmitter) by binding to GABA-A receptors. This increases chloride influx into neurons, producing anxiolytic, sedative, anticonvulsant and muscle-relaxant effects.
Directions for Use
Injection for use only under medical supervision. Given as slow IV injection/IV infusion or deep IM injection as prescribed; dose and rate depend on indication, age and clinical response. Avoid rapid IV administration; monitor breathing, blood pressure and level of sedation.
Schedule X medicine: use strictly on prescription and under supervision. Risk of respiratory depression, excessive sedation and hypotension—higher in elderly, debilitated patients, and with other CNS depressants. Use with caution in liver disease, COPD/sleep apnoea, myasthenia gravis, depression/suicidality. May cause dependence, tolerance and withdrawal; avoid abrupt discontinuation after repeated doses. Do not drive/operate machinery after receiving. IV use requires resuscitation facilities.

Drug Interactions
Alcohol and other CNS depressants (opioids, barbiturates, sedating antihistamines, antipsychotics) increase sedation/respiratory depression; opioids carry high risk of profound sedation/coma. CYP inhibitors (cimetidine, omeprazole, fluoxetine, fluvoxamine, azole antifungals, macrolides) may increase diazepam levels. CYP inducers (rifampicin, carbamazepine, phenytoin, phenobarbital) may reduce effect. Valproate may increase sedation. Theophylline/caffeine may reduce sedative effect.
