Monograph
Tablet
Treatment of HIV-1 infection as part of combination antiretroviral therapy (ART); prevention of mother-to-child transmission of HIV (as per specialist regimen/guidelines).
Zidovudine is a nucleoside reverse transcriptase inhibitor (NRTI). It is converted inside cells to an active triphosphate form that inhibits HIV reverse transcriptase and causes chain termination, reducing viral replication.
Tablet for oral use. Take exactly as prescribed, usually in combination with other antiretroviral medicines; may be taken with or without food. Take at the same time each day and do not stop without medical advice.
Nausea, vomiting, headache, diarrhea, abdominal pain, fatigue, anemia, neutropenia, fever, muscle pain, insomnia, rash
Use only under supervision of an HIV specialist and always as part of combination therapy. May cause bone marrow suppression (anemia, neutropenia); regular CBC monitoring required. Risk of lactic acidosis and severe hepatomegaly with steatosis; seek urgent care for persistent nausea/vomiting, abdominal pain, deep rapid breathing, or severe weakness. Use caution in liver disease and in patients with pre-existing anemia/neutropenia. Long-term use may be associated with myopathy and lipoatrophy. Do not share needles; continue safe sex practices as this medicine reduces viral load but does not cure HIV.
Other myelosuppressive drugs (e.g., ganciclovir, interferon alfa, ribavirin) may increase risk of anemia/neutropenia; probenecid may increase zidovudine levels; valproate may increase zidovudine exposure; rifampicin may reduce zidovudine concentrations; avoid/stay cautious with other nephrotoxic/hepatotoxic medicines; do not co-administer with stavudine (antagonistic effect).
Store below 25°C in a cool, dry place. Protect from moisture and light. Keep out of reach of children.
Zidovudine 300 mg Tablet, Zidovudine Syrup, Zidovudine + Lamivudine Tablet (150/300), Zidovudine + Lamivudine + Nevirapine Tablet, Zidovudine + Lamivudine + Efavirenz Tablet
