Glimepiride (1mg) + Metformin (1000mg) + Sitagliptin (50mg)
Type 2 diabetes mellitus in adults when diet and exercise alone are not sufficient; used to improve blood glucose control.
Glimepiride (a sulfonylurea) increases insulin release from the pancreas and improves peripheral glucose utilization. Metformin (a biguanide) reduces glucose production in the liver and improves insulin sensitivity, helping lower fasting and post‑meal blood sugar.
Take orally as prescribed, usually with/after meals to reduce stomach upset. Swallow tablet whole with water; do not crush unless advised. Monitor blood glucose regularly; dose is individualized based on control and tolerability.
Hypoglycaemia, nausea, vomiting, diarrhoea, abdominal pain, loss of appetite, metallic taste, headache, dizziness, weight gain
Risk of hypoglycaemia (especially with missed meals, alcohol, strenuous exercise, or with other antidiabetics); carry glucose source. Metformin can rarely cause lactic acidosis—risk increases with kidney impairment, dehydration, severe infection, heart/respiratory failure, or heavy alcohol use. Avoid/adjust in renal impairment; check eGFR before and during therapy. Temporarily stop metformin before iodinated contrast procedures or major surgery as advised. Use caution in liver disease; may cause vitamin B12 deficiency with long-term use. Not for type 1 diabetes or diabetic ketoacidosis.
Alcohol (increases hypoglycaemia/lactic acidosis risk), insulin/other antidiabetics (additive hypoglycaemia), beta-blockers (mask hypoglycaemia symptoms), corticosteroids/thiazides (raise glucose), ACE inhibitors (may enhance glucose-lowering), rifampicin (reduces sulfonylurea effect), azole antifungals/fluoroquinolones/salicylates (may increase hypoglycaemia), iodinated contrast media (lactic acidosis risk with metformin), cimetidine (raises metformin levels)
