Important Warnings

Highly teratogenic: contraindicated in pregnancy. Effective contraception is required for women of childbearing potential during treatment and for at least 3 years after stopping; regular pregnancy tests as advised. Do not donate blood during treatment and for 3 years after stopping. Avoid alcohol during treatment and for 2 months after stopping (can increase formation of etretinate and prolong teratogenic risk). Monitor liver function tests and lipid profile; use caution in liver disease, hyperlipidemia, diabetes, obesity. May cause mood changes, pseudotumor cerebri (severe headache/visual changes) and decreased night vision—avoid driving if affected. Avoid excessive sun/UV exposure.

Tetracyclines (risk of intracranial hypertension)
vitamin A or other retinoids (additive toxicity)
methotrexate (increased hepatotoxicity risk)
alcohol (increases etretinate formation)
phenytoin (protein binding interactions)
progestin-only “minipill” contraception may be less reliable—use effective contraception as advised.
