Treatment of type 2 diabetes mellitus in adults when diet, exercise and weight reduction alone are insufficient; helps control blood glucose levels.
Gliclazide is a sulfonylurea antidiabetic that stimulates insulin release from pancreatic beta cells by closing ATP-sensitive potassium channels. The modified-release formulation provides a prolonged effect to help maintain stable blood glucose control.
Oral tablet (modified release). Typically taken once daily with breakfast; swallow whole with water—do not crush, chew, or split. Use exactly as prescribed and monitor blood glucose regularly; avoid skipping meals.
Hypoglycaemia, dizziness, headache, nausea, abdominal pain, diarrhoea, constipation, weight gain, skin rash, itching
Risk of hypoglycaemia (higher with missed meals, heavy exercise, alcohol, elderly, renal/hepatic impairment); educate on recognizing and treating low sugar. Use with caution in kidney or liver disease; dose adjustment/alternative may be needed. Not recommended in type 1 diabetes, diabetic ketoacidosis, or severe infections/major surgery (may need insulin). Inform doctor about G6PD deficiency (risk of haemolysis). Avoid driving/operating machinery if symptoms of hypoglycaemia occur.
Other antidiabetics/insulin (increased hypoglycaemia risk), alcohol (may cause hypoglycaemia), beta-blockers (may mask hypoglycaemia symptoms), fluconazole/miconazole (increase gliclazide levels), macrolide antibiotics (e.g., clarithromycin), NSAIDs/salicylates (enhance hypoglycaemia), corticosteroids/thiazide diuretics (increase blood glucose), rifampicin (reduces effect), warfarin (may alter anticoagulant effect)
