Type 2 diabetes mellitus (non-insulin dependent) to improve blood glucose control along with diet and exercise
Gliclazide is a sulfonylurea that stimulates insulin release from pancreatic beta cells by closing ATP-sensitive potassium channels. This increases insulin secretion and helps lower blood glucose levels.
Take by mouth exactly as prescribed. Usually taken once daily (if modified release) or as directed; take with breakfast/food to reduce risk of low blood sugar. Swallow tablet whole with water; do not crush/chew if modified release.
Hypoglycaemia (low blood sugar), nausea, abdominal pain, diarrhoea, headache, dizziness, weight gain, skin rash
Risk of hypoglycaemia—more likely with missed meals, excessive exercise, alcohol intake, elderly patients, or kidney/liver impairment. Monitor blood glucose regularly. Use with caution in renal/hepatic disease, G6PD deficiency, malnutrition, and in patients with irregular eating habits. Stop and seek care for severe hypoglycaemia, jaundice, or allergic reactions. Not indicated for type 1 diabetes or diabetic ketoacidosis.
Other antidiabetics/insulin (increased hypoglycaemia risk), alcohol (may cause hypoglycaemia and disulfiram-like reaction), beta-blockers (mask hypoglycaemia symptoms), azole antifungals e.g., miconazole/fluconazole (increase effect), macrolides, NSAIDs/salicylates (increase effect), corticosteroids and thiazide diuretics (reduce glucose control), rifampicin (reduces effect), ACE inhibitors (may increase hypoglycaemia risk)
