Treatment of serious fungal infections such as invasive aspergillosis, serious Candida infections (including fluconazole-resistant), and other invasive fungal infections as per physician assessment
Voriconazole is a triazole antifungal that inhibits fungal cytochrome P450–dependent 14-α-sterol demethylase. This blocks ergosterol synthesis, leading to defective fungal cell membrane formation and inhibition of fungal growth.
For intravenous use only. Reconstitute and dilute as directed in the product label and administer as IV infusion by a healthcare professional. Dose and duration depend on indication, body weight, liver function and clinical response; may be given with or without food (IV route).
Visual disturbances (blurred vision, photophobia, color vision changes), fever, nausea, vomiting, diarrhea, abdominal pain, headache, rash, liver enzyme elevation, infusion-site reactions
Prescription-only medicine; IV administration under medical supervision. Monitor liver function (risk of hepatitis/hepatic failure) and discontinue if significant toxicity occurs. Can cause visual disturbances and photosensitivity/phototoxicity; avoid excessive sunlight and use protection. Risk of QT prolongation/arrhythmias—use caution in patients with cardiac disease, electrolyte abnormalities, or when combined with QT-prolonging drugs. Use caution and dose adjustment/monitoring in hepatic impairment; monitor renal function (IV formulation excipients may affect patients with renal impairment).
Strong interaction potential via CYP3A4/CYP2C19/CYP2C9: contraindicated/avoid with rifampicin, carbamazepine, long-term phenytoin/high-dose ritonavir, St John’s wort; may increase levels of warfarin, cyclosporine, tacrolimus, sirolimus, certain statins, benzodiazepines, sulfonylureas; caution with QT-prolonging medicines and drugs affecting electrolytes
