Important Warnings

Do not use if allergic to esomeprazole/domperidone or other PPIs. Domperidone may increase risk of QT prolongation/serious heart rhythm problems—use caution in heart disease, electrolyte imbalance, and in older patients; avoid with known prolonged QT. Prolonged PPI use may be associated with low magnesium, vitamin B12 deficiency, fractures, and C. difficile-associated diarrhea; seek care for severe/persistent diarrhea. May mask symptoms of gastric cancer—consult doctor for alarm symptoms (weight loss, vomiting blood, black stools, difficulty swallowing). Use caution in severe liver disease; kidney disease: consult doctor.

QT-prolonging drugs (e.g.

sotalol
certain antipsychotics/antidepressants)
strong CYP3A4 inhibitors (e.g.
ketoconazole/itraconazole
erythromycin/clarithromycin
ritonavir) increase domperidone levels and arrhythmia risk; warfarin (monitor INR)
clopidogrel (reduced activation possible with some PPIs)
digoxin
methotrexate (high dose)
atazanavir/nelfinavir (reduced absorption with PPIs)
iron salts and other drugs needing acidic pH (reduced absorption)
