Moderate to severe acute pain requiring opioid analgesic treatment (e.g., musculoskeletal pain, postoperative pain) when other analgesics are inadequate.
Tapentadol is a centrally acting opioid analgesic. It works by dual mechanisms: μ-opioid receptor agonism and inhibition of noradrenaline reuptake in the CNS, reducing pain signal transmission and perception.
Take exactly as prescribed by a doctor. Swallow tablet whole with water; can be taken with or without food. Do not crush/chew. Avoid driving or operating machinery after taking; dose and duration depend on pain severity.
Nausea, vomiting, dizziness, drowsiness/somnolence, constipation, headache, dry mouth, fatigue
Prescription-only opioid; risk of dependence, abuse and withdrawal. May cause severe drowsiness, respiratory depression and low blood pressure—higher risk in elderly, debilitated, or with lung disease (asthma/COPD). Avoid combining with other CNS depressants. Use caution in head injury/raised intracranial pressure, seizure disorders, liver/kidney impairment. Do not stop abruptly after prolonged use. Not recommended in severe respiratory depression or paralytic ileus. Keep out of reach of children.
Alcohol, benzodiazepines and other sedatives (additive CNS/respiratory depression), other opioids, antihistamines/sedating drugs, MAO inhibitors (avoid; risk of serious reactions), serotonergic drugs (SSRIs/SNRIs/TCAs/triptans/linezolid) may increase serotonin syndrome risk, strong CYP2D6/CYP3A4 modulators may alter exposure (clinical significance variable)
