Pregabalin 150 mg (sustained release)
Neuropathic (nerve) pain due to diabetes, shingles (post-herpetic neuralgia) or spinal cord injury; fibromyalgia; adjunct treatment of partial-onset seizures (as add-on therapy)
Pregabalin binds to the alpha-2-delta subunit of voltage-gated calcium channels in the CNS. This reduces release of excitatory neurotransmitters and decreases abnormal nerve signaling, relieving neuropathic pain and helping control seizures.
Oral sustained-release tablet. Swallow whole with water; do not crush, break or chew. Take once daily as prescribed, preferably at the same time each day; may be taken with or without food. Do not stop suddenly—taper as advised by the doctor.
Dizziness, sleepiness (somnolence), blurred vision, dry mouth, weight gain, peripheral edema (swelling), fatigue, headache, constipation, difficulty concentrating
May cause dizziness/drowsiness—avoid driving or operating machinery until effects are known. Use with caution in kidney disease (dose adjustment required). Can cause swelling/weight gain; caution in heart failure. Rare risk of allergic reactions/angioedema and vision changes—seek medical help if swelling of face/lips, rash, or breathing difficulty occurs. May increase suicidal thoughts in some patients—monitor mood/behavior changes. Do not discontinue abruptly to avoid withdrawal symptoms.
Alcohol and other CNS depressants (opioids, benzodiazepines, sedating antihistamines) may increase drowsiness/respiratory depression risk; Opioids (e.g., oxycodone, tramadol) increase sedation and constipation risk; ACE inhibitors may increase risk of angioedema; Antidiabetic thiazolidinediones (pioglitazone, rosiglitazone) may increase edema/weight gain
