Monograph
Capsule
Gastroesophageal reflux disease (GERD), acidity/heartburn, gastritis, peptic ulcer-related acid symptoms, nausea and vomiting associated with reflux
Pantoprazole is a proton pump inhibitor that reduces stomach acid by blocking the H+/K+ ATPase pump in gastric parietal cells. Domperidone is a dopamine (D2) receptor antagonist that improves gastric motility and helps relieve nausea by enhancing stomach emptying.
Take as directed by a doctor. Commonly taken as 1 capsule once daily, 30 minutes before breakfast; swallow whole without crushing/chewing (SR/EC formulation).
Headache, diarrhea, abdominal pain, nausea, dry mouth, dizziness, flatulence, constipation
Use with caution in liver disease. Domperidone may increase risk of QT prolongation/serious heart rhythm problems—avoid in known prolonged QT, significant electrolyte disturbances, or moderate to severe heart disease. Seek medical advice if black stools, vomiting blood, persistent symptoms, or unexplained weight loss occur. Long-term PPI use may be associated with low magnesium, vitamin B12 deficiency, fractures, and C. difficile diarrhea.
QT-prolonging drugs (e.g., amiodarone, sotalol, certain antipsychotics), strong CYP3A4 inhibitors (ketoconazole, itraconazole, clarithromycin, erythromycin, ritonavir) may increase domperidone levels; anticoagulants like warfarin (monitor), drugs needing acidic pH for absorption (ketoconazole/itraconazole, atazanavir), methotrexate (high dose), clopidogrel (possible reduced activation with some PPIs)
