Treatment of type 2 diabetes mellitus to improve glycaemic control as an adjunct to diet and exercise; can be used alone or in combination with other antidiabetic medicines as prescribed.
Sitagliptin is a DPP-4 inhibitor that increases levels of incretin hormones (GLP-1 and GIP). This enhances glucose-dependent insulin release and reduces glucagon secretion, lowering blood glucose with a low risk of hypoglycaemia when used alone.
Take by mouth as directed by the doctor, usually once daily. Can be taken with or without food; swallow tablet with water at the same time each day. Dose adjustment may be needed in kidney disease; do not stop or change dose without medical advice.
Headache, nasopharyngitis, upper respiratory tract infection, nausea, diarrhoea, abdominal pain
Not for type 1 diabetes or diabetic ketoacidosis. Use with caution in kidney impairment (dose adjustment required) and in elderly; monitor renal function. Seek urgent care for severe abdominal pain (possible pancreatitis), signs of serious allergic reaction/angioedema, severe joint pain, or blistering rash (bullous pemphigoid). Hypoglycaemia risk increases when combined with insulin or sulfonylureas; dose adjustment of those agents may be needed.
Insulin and sulfonylureas (increased hypoglycaemia risk), digoxin (may increase digoxin levels; monitor), strong P-gp/CYP3A4 inhibitors/inducers (clinically limited effect but caution), alcohol (may worsen glycaemic control)
