Type 2 diabetes mellitus: to improve glycaemic control as an adjunct to diet and exercise; may be used alone or in combination with other antidiabetic medicines (as prescribed).
Sitagliptin is a DPP-4 inhibitor that increases levels of incretin hormones (GLP-1 and GIP). This enhances glucose‑dependent insulin release and reduces glucagon secretion, thereby lowering blood glucose with a low risk of hypoglycaemia when used alone.
Take by mouth exactly as prescribed, usually once daily, with or without food. Swallow the tablet whole with water; take at the same time each day. Dose adjustment may be needed in kidney disease—follow doctor’s advice.
Headache, nasopharyngitis (common cold), sore throat, upper respiratory tract infection, diarrhoea, nausea, stomach discomfort
Not for type 1 diabetes or diabetic ketoacidosis. Use with caution in kidney impairment (dose adjustment required); periodic renal function monitoring recommended. Seek medical help if severe abdominal pain occurs (possible pancreatitis) or if signs of serious allergy occur (rash, swelling, breathing difficulty). May cause hypoglycaemia when combined with insulin or sulfonylureas—dose adjustments may be needed. Report severe joint pain or blistering skin rash (bullous pemphigoid).
Insulin and sulfonylureas (increased risk of hypoglycaemia), digoxin (may increase digoxin levels—monitor), strong CYP3A4/P-gp inhibitors/inducers have limited effect but use clinical monitoring; other antidiabetics may require dose adjustments.
Store below 25°C in a cool, dry place. Protect from moisture and direct light. Keep out of reach of children.
