Valproate increases the level of the inhibitory neurotransmitter GABA in the brain and also reduces excessive neuronal firing by modulating sodium and calcium channels. This stabilizes electrical activity in the brain and helps prevent seizures and mood episodes.

Oral prolonged/controlled release tablet: swallow whole with water; do not crush or chew. Usually taken once or twice daily as prescribed, preferably at the same time each day; take with food if gastric upset occurs. Do not stop suddenly without medical advice.

Prescription-only medicine. Risk of severe liver injury (especially in children <2 years or with metabolic disorders); seek help for persistent vomiting, abdominal pain, jaundice or dark urine. Can cause pancreatitis (severe abdominal pain), hyperammonemia/encephalopathy, thrombocytopenia/bleeding, and sedation. Monitor liver function tests and blood counts as advised. Not recommended in pregnancy due to high risk of birth defects and neurodevelopmental disorders; women of childbearing potential should use effective contraception and consult doctor. Use caution in hepatic impairment, urea cycle disorders, mitochondrial disorders, and elderly. May impair driving/operating machinery.

Carbamazepine, phenytoin, phenobarbital/primidone, lamotrigine (increased risk of severe rash), topiramate (hyperammonemia), clobazam, warfarin/other anticoagulants (bleeding risk), aspirin/NSAIDs (increased free valproate/bleeding), rifampicin, erythromycin, CNS depressants (additive sedation)

Store below 25°C in a dry place, protected from light and moisture. Keep in original blister pack and out of reach of children.
