Amicobal Pg Tablet contains Amitriptyline (10mg) + Pregabalin (75mg). It belongs to the Antidepressant and Gabapentinoid class of medicines.

How Amicobal Pg Tablet works

Amicobal PG 10mg/75mg Tablet is a combination of two medicines: Amitriptyline and Pregabalin. Amitriptyline is a tricyclic antidepressant which increases the levels of chemical messengers (serotonin and noradrenaline) that stop the movement of pain signals in the brain. Together, they relieve neuropathic pain (pain from damaged nerves). Pregabalin is an alpha 2 delta ligand which decreases pain by modulating calcium channel activity of the nerve cells.

How to take Amicobal Pg Tablet

Take this medicine in the dose and duration as advised by your doctor. Swallow it as a whole. Do not chew, crush or break it. Amicobal PG 10mg/75mg Tablet may be taken with or without food.

Take it exactly as your doctor prescribed. Do not change the dose or stop early on your own.

Missed dose and overdose

If you miss a dose

If you miss a dose of Amicobal PG 10mg/75mg Tablet, take it as soon as possible. However, if it is almost time for your next dose, skip the missed dose and go back to your regular schedule. Do not double the dose.

If you take too much

An overdose of this medicine can cause fits, dangerous heart rhythm disturbance and coma, and is particularly dangerous with the older tricyclic medicines. It can cause deep drowsiness, confusion, unsteadiness and slowed breathing, especially with an opioid or alcohol. Do not wait for symptoms to appear — go to the nearest emergency department and take the pack or strip with you.

Most side effects of Amicobal Pg are mild and settle as the body adjusts. Reported side effects include:

Stop the medicine and seek help immediately for a rash, swelling of the face or lips, wheezing or difficulty breathing. Tell your doctor about any side effect that persists or worries you.

Safety advice for Amicobal Pg Tablet

Warnings and precautions

These medicines take two to four weeks to begin working, and longer for the full effect. Stopping at two weeks because nothing has changed is the commonest mistake. Never stop suddenly. Abrupt withdrawal causes dizziness, electric-shock sensations, irritability and flu-like symptoms — your doctor will reduce the dose gradually. Tell your doctor or go to hospital immediately if thoughts of harming yourself appear or worsen, particularly in the first weeks or after a dose change, and especially in anyone under twenty-five. Seek urgent care for agitation with fever, shivering, a racing heart, muscle twitching or confusion, particularly if more than one medicine affecting serotonin is being taken. Side effects are usually worst in the first fortnight and then settle. This medicine causes drowsiness and unsteadiness, particularly when you start it or the dose goes up. Do not drive or operate machinery until you know how it affects you. Never stop it suddenly. Stopping abruptly can cause anxiety, sleeplessness, sweating and, in people taking it for epilepsy, fits. Your doctor will taper it. Tell your doctor about any change in mood or any thoughts of harming yourself. The dose is usually built up slowly from a low starting point — follow the schedule your doctor gave you rather than jumping to the full dose. Pregabalin comes under Schedule H1 from 16 November 2026 and will then need a prescription and a pharmacy register entry.

Do not take Amicobal Pg if

Not for anyone with a known allergy to this medicine or to any ingredient in the pack. Must not be combined with a monoamine oxidase inhibitor, or started within the gap your doctor specifies after one.

Interactions with other medicines

Tramadol, triptans for migraine, linezolid, St John's wort and other antidepressants can together raise serotonin dangerously. Anti-inflammatory painkillers and blood thinners increase the bleeding risk when combined with this group. Opioid painkillers, alcohol, sleeping tablets and antihistamines all add to the drowsiness, and the combination with opioids can dangerously slow breathing. Antacids reduce the absorption of gabapentin — separate them by a couple of hours.

Store below 30°C in the original pack, away from direct sunlight and moisture. Keep out of the reach of children.

Amicobal PG 10mg/75mg Tablet is used to treat certain types of long-lasting pain caused by damage to nerves. It may ease the pain within a few days but it may take 2-3 weeks before you see maximum benefit. Inform your doctor if you are taking any other pain-relieving medicines. Along with taking this medicine, your doctor might advise you to undergo physiotherapy to get relief from pain. Amicobal PG 10mg/75mg Tablet might cause dry mouth. Frequent sips of water may help ease the discomfort.
