Treatment of fungal infections such as aspergillosis, blastomycosis, histoplasmosis, onychomycosis (nail fungus), tinea (ringworm), candidiasis and other susceptible systemic or superficial mycoses

Itraconazole is a triazole antifungal that inhibits fungal CYP450-dependent synthesis of ergosterol, an essential component of the fungal cell membrane. This disrupts membrane integrity and stops fungal growth and replication.

Take as directed by a doctor. Capsules are usually taken with food for better absorption; swallow whole with water. Take at the same time each day and complete the full course even if symptoms improve.

Not recommended in patients with heart failure/ventricular dysfunction; seek medical advice if shortness of breath, swelling or sudden weight gain occurs. Use with caution in liver disease; liver function monitoring may be needed and stop if symptoms of hepatitis (fatigue, loss of appetite, dark urine, jaundice) appear. Avoid use in pregnancy unless clearly needed; effective contraception advised during treatment and for some time after. Many significant drug interactions—inform your doctor about all medicines. Acid-reducing agents can lower absorption; take as advised.

Contraindicated/major interactions with simvastatin/lovastatin, quinidine, dofetilide, dronedarone, ergot alkaloids, oral midazolam/triazolam, certain calcium channel blockers, some antiarrhythmics. Increases levels of warfarin, digoxin, cyclosporine/tacrolimus, certain benzodiazepines, sulfonylureas. Rifampicin/rifabutin, phenytoin, carbamazepine, St John’s wort reduce itraconazole levels. PPIs/H2 blockers/antacids may reduce absorption of capsules.

Store below 25°C in a cool, dry place away from moisture and direct light. Keep in original pack and out of reach of children.
