Treatment of fungal infections such as aspergillosis, histoplasmosis, blastomycosis, onychomycosis (nail fungus), tinea infections (ringworm), oral/vaginal candidiasis, and other susceptible systemic or superficial mycoses.

Itraconazole is a triazole antifungal that inhibits fungal CYP450-dependent 14-α-demethylase, reducing ergosterol synthesis. This disrupts the fungal cell membrane and inhibits growth and survival of the fungus.

Capsule: take exactly as prescribed. Usually taken with food (preferably a full meal) to improve absorption; swallow whole with water. Take at the same time each day and complete the full course; do not stop early even if symptoms improve.

Nausea, abdominal pain, diarrhea, headache, dizziness, rash, liver enzyme increase

Prescription-only antifungal. Can cause serious liver problems—seek care for jaundice, dark urine, persistent nausea/vomiting. Not recommended in patients with congestive heart failure/ventricular dysfunction (may worsen heart failure). Many significant drug interactions; inform doctor about all medicines. Use caution in liver disease; monitoring of liver function may be required. May cause dizziness—avoid driving if affected.

Strong interactions with drugs affecting CYP3A4. Contraindicated/avoid with: simvastatin/lovastatin, oral midazolam/triazolam, quinidine, dofetilide, cisapride, pimozide, ergot alkaloids. Levels affected by rifampicin/rifabutin, phenytoin, carbamazepine (reduce itraconazole). Increases levels of: warfarin, certain calcium channel blockers, digoxin, cyclosporine/tacrolimus, some benzodiazepines, certain antiarrhythmics. Reduced absorption with antacids, H2 blockers, PPIs—separate dosing and follow medical advice.

Store below 25°C in a cool, dry place. Protect from moisture and direct light. Keep in original pack and out of reach of children.
