Monograph
Capsule
Gastroesophageal reflux disease (GERD), acidity/heartburn, indigestion (dyspepsia), nausea and vomiting associated with acid reflux
Pantoprazole is a proton pump inhibitor that reduces stomach acid by blocking the gastric H+/K+ ATPase pump. Domperidone is a peripheral dopamine (D2) receptor antagonist that increases gastric motility and helps relieve nausea/vomiting, improving reflux symptoms.
Take 1 capsule as prescribed, usually once daily 30 minutes before food (preferably before breakfast). Swallow whole; do not crush or chew (contains enteric-coated/SR components).
Headache, diarrhea, abdominal pain, nausea, dry mouth, dizziness, flatulence, constipation
Use with caution in liver disease. Domperidone may increase risk of QT prolongation/serious heart rhythm problems—avoid in patients with known QT prolongation, significant cardiac disease, electrolyte disturbances, or when used with QT-prolonging medicines. Seek medical help for black stools, blood in vomit, persistent vomiting, unexplained weight loss, or severe abdominal pain. Long-term PPI use may be associated with low magnesium/vitamin B12, bone fracture risk, and gastrointestinal infections; use the lowest effective dose.
Ketoconazole/itraconazole/posaconazole/erlotinib (reduced absorption with pantoprazole), warfarin (monitor INR), digoxin (risk of hypomagnesemia-related toxicity), methotrexate high dose (increased levels), clopidogrel (possible reduced activation; consider alternative PPI if needed), strong CYP3A4 inhibitors like clarithromycin/erythromycin/ketoconazole/ritonavir (increase domperidone levels and QT risk), other QT-prolonging drugs (e.g., amiodarone, sotalol, quinolones, macrolides, antipsychotics)
