How It Works
Domperidone is a dopamine (D2) receptor antagonist with prokinetic and antiemetic action. It increases gastric motility and accelerates stomach emptying, and blocks dopamine receptors in the chemoreceptor trigger zone to reduce nausea and vomiting.
Directions for Use
Oral suspension. Shake well before use. Take the prescribed dose using a measuring cup/spoon, usually 15–30 minutes before meals; may be taken before bedtime if advised. Dose and duration should be as directed by the doctor (especially in children).
Prescription (Schedule H) medicine. Do not use if you have known allergy to domperidone, gastrointestinal bleeding/obstruction/perforation, or prolactin-secreting pituitary tumor (prolactinoma). Use with caution in liver impairment; dose adjustment/avoidance may be needed. Domperidone may prolong QT interval and cause serious heart rhythm problems; use lowest effective dose for shortest duration, especially in older adults or those with heart disease, electrolyte imbalance, or taking QT-prolonging medicines. Seek urgent care for palpitations, fainting, or irregular heartbeat.

Strong CYP3A4 inhibitors (ketoconazole, itraconazole, fluconazole, clarithromycin, erythromycin, ritonavir) increase domperidone levels and QT risk; other QT-prolonging drugs (amiodarone, sotalol, quinolones, antipsychotics) increase arrhythmia risk; anticholinergics may reduce prokinetic effect; antacids/acid suppressants may reduce absorption if taken together (separate dosing).
