Gastroesophageal reflux disease (GERD) and heartburn, erosive esophagitis, peptic ulcer disease, prevention of NSAID-associated ulcers, Helicobacter pylori eradication (with antibiotics), Zollinger–Ellison syndrome
Esomeprazole is a proton pump inhibitor (PPI). It irreversibly blocks the gastric H+/K+ ATPase (proton pump) in stomach parietal cells, reducing acid secretion and allowing ulcers/erosions to heal.
Tablet: take as prescribed, usually once daily 30–60 minutes before food (preferably before breakfast). Swallow whole with water; do not crush or chew. Duration depends on condition (often 2–8 weeks for GERD/ulcers).
Headache, nausea, diarrhea, abdominal pain, flatulence, constipation, dizziness
Use with caution in severe liver disease. Long-term use may increase risk of vitamin B12 deficiency, low magnesium, bone fractures, and intestinal infections (e.g., C. difficile). Seek medical advice if alarm symptoms occur (unexplained weight loss, persistent vomiting, GI bleeding, difficulty swallowing). May mask symptoms of gastric malignancy; evaluation may be needed if symptoms persist.
Warfarin/other anticoagulants (monitor INR), clopidogrel (reduced activation), diazepam, phenytoin, digoxin, methotrexate (high dose), tacrolimus, ketoconazole/itraconazole (reduced absorption), atazanavir/nelfinavir (avoid), rifampicin and St John’s wort (may reduce effect)
