Isotretinoin is an oral retinoid that reduces sebaceous gland size and sebum production. It also normalizes follicular keratinization and has anti-inflammatory effects, thereby reducing acne lesions.
Oral capsule; take exactly as prescribed by a dermatologist. Usually taken once or twice daily with food (preferably with a fatty meal) to improve absorption; swallow whole with water, do not crush/chew. Follow the full course and regular follow-ups/lab tests.
Dry lips, dry skin, dry eyes, nosebleeds, skin peeling, photosensitivity, headache, muscle/joint pain, increased liver enzymes, increased triglycerides, mood changes
Highly teratogenic: contraindicated in pregnancy and during breastfeeding; strict pregnancy prevention program required (effective contraception, pregnancy tests before/during/after therapy). Do not donate blood during treatment and for at least 1 month after stopping. Monitor liver function and lipids; use caution in history of depression, diabetes, hyperlipidaemia, liver disease. Avoid excessive sun/UV exposure; use sunscreen. Avoid vitamin A supplements and tetracycline antibiotics (risk of raised intracranial pressure). May reduce night vision; caution while driving at night. Do not use for mild/moderate acne unless advised.
Tetracyclines (e.g., doxycycline/minocycline), vitamin A/retinoid supplements, alcohol (worsens hypertriglyceridaemia/hepatotoxicity risk), hepatotoxic medicines, corticosteroids (bone effects), phenytoin (bone effects), St John’s wort (may reduce contraceptive efficacy)
Store below 25°C in a cool, dry place, protected from light and moisture. Keep in original blister pack; keep out of reach of children.
