Treatment of chronic hepatitis C virus (HCV) infection (used in combination with other antivirals as per genotype and doctor’s prescription).
Paritaprevir is an HCV NS3/4A protease inhibitor that blocks viral polyprotein processing and replication. Ritonavir acts mainly as a pharmacokinetic booster by inhibiting CYP3A4, increasing paritaprevir levels and effectiveness.
Take orally as prescribed by your doctor, usually with food. Swallow the capsule whole with water; do not crush or open. Take at the same time each day and complete the full course; do not stop without medical advice.
Nausea,diarrhoea,headache,fatigue,insomnia,itching,rash,raised liver enzymes
Prescription-only antiviral; use only under specialist supervision. Not recommended in severe hepatic impairment; monitor liver function before and during treatment. Risk of hepatitis B reactivation—HBV screening recommended before starting. Numerous clinically significant drug interactions due to ritonavir; do not start/stop other medicines without medical advice. Seek help for jaundice, dark urine, severe abdominal pain, persistent vomiting, severe rash or breathing difficulty.
Strong interactions with CYP3A substrates/inducers: rifampicin, carbamazepine, phenytoin, phenobarbital, St John’s wort (reduce effect/contraindicated); simvastatin/lovastatin (contraindicated), certain antiarrhythmics (e.g., amiodarone), sedatives (midazolam/triazolam), ergot derivatives, some antiretrovirals, immunosuppressants (tacrolimus/cyclosporine), anticoagulants (warfarin/DOACs—monitor), oral contraceptives containing ethinyl estradiol (avoid/alternative contraception).
