Treatment and prevention of vitamin D deficiency, vitamin D insufficiency, rickets/osteomalacia, adjunct in osteoporosis and low bone mineral density, supplementation in malabsorption or limited sun exposure
Cholecalciferol (vitamin D3) is converted in the liver and kidneys to active vitamin D (calcitriol). It increases intestinal absorption of calcium and phosphate and helps maintain normal blood calcium levels, supporting bone mineralization and muscle function.
Capsule for oral use. Commonly taken as 1 capsule (60000 IU) once weekly for 6–8 weeks or as advised by the doctor, then maintenance dosing as prescribed (often monthly). Take with food (preferably a meal containing fat) to enhance absorption; swallow whole with water.
Nausea, vomiting, constipation, abdominal pain, loss of appetite, headache, weakness, increased thirst, increased urination
Do not use in hypercalcemia, hypervitaminosis D, or history of vitamin D toxicity. Use with caution in kidney disease, kidney stones, sarcoidosis or other granulomatous disorders, and in patients on high calcium intake/supplements. Avoid exceeding prescribed dose; prolonged high dosing can cause toxicity (confusion, severe vomiting, dehydration, arrhythmias). Monitoring of calcium, phosphate and vitamin D levels may be required during high-dose therapy.
Thiazide diuretics (risk of hypercalcemia), digoxin/cardiac glycosides (arrhythmia risk with hypercalcemia), anticonvulsants like phenytoin/phenobarbital/carbamazepine (reduced vitamin D effect), rifampicin (reduced effect), glucocorticoids (reduced effect), cholestyramine/orlistat (reduced absorption), aluminum- or magnesium-containing antacids in renal impairment (risk of toxicity), high-dose calcium supplements (hypercalcemia risk)
