Chronic weight management (obesity/overweight) as an adjunct to reduced-calorie diet and increased physical activity in eligible adults; may be used when weight-related comorbidities are present as per physician advice.

Semaglutide is a GLP-1 receptor agonist. It increases satiety, reduces hunger and energy intake, and slows gastric emptying, leading to weight loss. It also improves glucose regulation by enhancing glucose-dependent insulin secretion and lowering glucagon.

Subcutaneous injection only (abdomen, thigh, or upper arm) once weekly on the same day each week, with or without food. Rotate injection sites; do not inject intravenously or intramuscularly. Follow the dose-escalation schedule prescribed by the doctor; if a dose is missed, take within 5 days otherwise skip and take next scheduled dose.

Prescription-only medicine. Risk of pancreatitis—seek medical help for severe persistent abdominal pain. May cause gallbladder disease (gallstones), dehydration and acute kidney injury especially with vomiting/diarrhoea; maintain fluids. Can increase heart rate; use caution in patients with cardiac disease. Risk of hypoglycaemia when used with insulin or sulfonylureas. Not recommended in patients with personal/family history of medullary thyroid carcinoma or MEN2; report neck lump/hoarseness/dysphagia. Use caution in severe gastrointestinal disease (e.g., gastroparesis). Do not use for type 1 diabetes or for diabetic ketoacidosis.
