Composition of Mezolam Injection

Mezolam Injection contains Midazolam 1 mg/mL (10 mL vial). It belongs to the Benzodiazepine (short-acting) class of medicines.

How Mezolam Injection works

Midazolam is a benzodiazepine that enhances the effect of GABA at the GABA-A receptor in the brain. This increases inhibitory neurotransmission, producing sedation, anxiolysis, muscle relaxation and anticonvulsant effects.

Side effects of Mezolam Injection

Most side effects of Mezolam are mild and settle as the body adjusts. Reported side effects include:

• Drowsiness
• sedation
• dizziness
• confusion
• amnesia
• low blood pressure
• slow/irregular breathing
• apnea
• nausea
• vomiting
• injection site pain

Stop the medicine and seek help immediately for a rash, swelling of the face or lips, wheezing or difficulty breathing. Tell your doctor about any side effect that persists or worries you.

Warnings and precautions

Schedule H1 medicine: use only on prescription under medical supervision. Can cause profound sedation, respiratory depression, apnea and hypotension—higher risk with opioids, alcohol, other sedatives, elderly, debilitated patients, sleep apnea, COPD or severe illness. Use caution in liver impairment; effects may be prolonged. Risk of paradoxical reactions (agitation, aggression) and anterograde amnesia. Potential for dependence/withdrawal with prolonged or repeated use; taper if used for extended periods. Not recommended to self-administer.

Do not take Mezolam if

Not for anyone with a known allergy to this medicine or to any ingredient in the pack. Not for people with severe breathing problems, sleep apnoea, or severe liver disease. Not for anyone with a history of dependence, except under specialist supervision.

Interactions with other medicines

Opioids (morphine,fentanyl,codeine,tramadol) and other CNS depressants (alcohol,antihistamines,antipsychotics,sedative antidepressants) increase respiratory depression. CYP3A4 inhibitors (ketoconazole,itraconazole,fluconazole,clarithromycin,erythromycin,ritonavir/cobicistat,grapefruit) increase midazolam levels; CYP3A4 inducers (rifampicin,carbamazepine,phenytoin,phenobarbital,St John’s wort) reduce effect. Caution with antihypertensives due to hypotension.

How to store Mezolam Injection

Store below 25°C in original carton, protected from light. Do not freeze. Keep out of reach of children.

Use it for the shortest course, and ask your doctor about talking therapy and sleep routine, which work without dependence. Take it only at bedtime if it is for sleep, and allow a full night before driving. Never share these medicines, and keep them secured.
