Monograph
Tablet
Treatment of hypertension (high blood pressure), including patients not adequately controlled on single therapy; helps reduce risk of cardiovascular events associated with hypertension.
Amlodipine is a calcium channel blocker that relaxes blood vessels by reducing calcium entry into vascular smooth muscle. Losartan is an angiotensin II receptor blocker (ARB) that blocks AT1 receptors, leading to vasodilation and reduced aldosterone effects; together they lower blood pressure effectively.
Take orally as prescribed, usually once daily at the same time each day, with or without food. Swallow whole with water; do not stop suddenly without medical advice. Dose adjustments may be needed based on BP response and kidney function.
Dizziness, headache, peripheral edema (swelling of ankles/feet), fatigue, flushing, nausea, palpitations, low blood pressure
Use with caution in kidney impairment, liver disease, dehydration/low salt intake, and in elderly due to risk of hypotension. Monitor blood pressure, kidney function and serum potassium. Avoid in pregnancy (especially 2nd/3rd trimester) due to risk of fetal harm from losartan; discontinue if pregnancy is detected. May cause dizziness—avoid driving if affected.
Potassium supplements/potassium-sparing diuretics (risk of hyperkalemia), NSAIDs (reduced antihypertensive effect and possible kidney harm), other antihypertensives/diuretics (additive hypotension), lithium (increased lithium levels/toxicity), CYP3A4 inhibitors/inducers affecting amlodipine levels (e.g., clarithromycin, azoles, rifampicin), alcohol (may increase dizziness)
Store below 25°C in a dry place, protected from light and moisture. Keep out of reach of children.
