Hormone replacement therapy (HRT) for symptoms of menopause (hot flashes, night sweats, vaginal dryness) and prevention of postmenopausal osteoporosis when appropriate; treatment of estrogen deficiency (e.g., due to ovarian failure or after oophorectomy) as advised by a doctor.
Estradiol is a natural estrogen hormone. It replaces or supplements estrogen levels that fall after menopause or due to ovarian insufficiency, helping relieve vasomotor and urogenital symptoms and maintaining bone mineral density. It acts by binding to estrogen receptors and regulating gene expression in target tissues.
Take by mouth as prescribed, usually once daily at the same time each day. Can be taken with or without food. Swallow with water; do not exceed the prescribed dose. HRT regimens may be cyclic or continuous; if you have a uterus, a progestogen is often added to reduce endometrial cancer risk.

Prescription-only hormone. Increased risk of blood clots (DVT/PE), stroke and heart attack (especially with risk factors such as smoking, obesity, immobility, thrombophilia). May increase risk of breast cancer with long-term use and risk of endometrial hyperplasia/cancer if used without progestogen in women with an intact uterus. Avoid/use only under specialist advice in history of breast/estrogen-dependent cancer, unexplained vaginal bleeding, active or past VTE, severe liver disease, porphyria, or pregnancy. Stop and seek urgent care for chest pain, sudden breathlessness, severe headache, vision changes, leg swelling/pain, or jaundice. Regular follow-up (breast exam/mammography as advised, BP, and evaluation of abnormal bleeding).
CYP3A4 inducers may reduce effect (rifampicin, rifabutin, carbamazepine, phenytoin, phenobarbital, primidone, St John’s wort); CYP3A4 inhibitors may increase levels (ketoconazole/itraconazole, erythromycin/clarithromycin, ritonavir/cobicistat); may affect thyroid hormone requirements (increase TBG—monitor if on levothyroxine); may alter effects of anticoagulants and corticosteroids—monitor.
