Monograph
Glyceryl trinitrate (Nitroglycerin) 6.4 mg modified-release/extended-release tablet
Prevention (prophylaxis) of angina pectoris (chest pain) due to coronary artery disease; used to reduce frequency of angina attacks (not for immediate relief of an acute attack).
Nitroglycerin is converted to nitric oxide in vascular smooth muscle, which relaxes and dilates veins and arteries. This reduces preload and myocardial oxygen demand and improves coronary blood flow, helping prevent angina.
Take by mouth exactly as prescribed. Swallow the extended-release tablet whole with water; do not crush or chew. Usually taken 1–2 times daily; often a daily nitrate-free interval may be advised to prevent tolerance. Can be taken with or without food; take at the same times each day.
Headache, dizziness, flushing, lightheadedness, low blood pressure, nausea, weakness, palpitations
Do not use with PDE-5 inhibitors (sildenafil, tadalafil, vardenafil) or riociguat due to risk of severe hypotension. May cause dizziness/fainting—rise slowly and use caution while driving. Use caution in low blood pressure, volume depletion, severe anemia, increased intracranial pressure, hypertrophic obstructive cardiomyopathy, recent heart attack/heart failure, and hepatic/renal impairment. Do not stop abruptly without medical advice; tolerance can develop—follow prescribed nitrate-free interval.
PDE-5 inhibitors (sildenafil/tadalafil/vardenafil), riociguat, other antihypertensives, alcohol, diuretics, beta-blockers/calcium channel blockers (additive hypotension), ergot derivatives (reduced antianginal effect), tricyclic antidepressants/antipsychotics (additive orthostatic hypotension)
Store at room temperature (below 25°C), protected from moisture and light, in the original tightly closed container. Keep out of reach of children.
