Monograph
Tablet
Management of high cholesterol (hypercholesterolaemia, mixed dyslipidaemia), reduction of elevated LDL-C and triglycerides, prevention of cardiovascular events (heart attack, stroke) in high-risk patients
Atorvastatin inhibits HMG‑CoA reductase, the key enzyme in hepatic cholesterol synthesis. This lowers LDL cholesterol and triglycerides and modestly increases HDL, reducing atherosclerotic plaque progression and cardiovascular risk.
Oral tablet. Take once daily at the same time each day, with or without food, as prescribed. Continue diet and lifestyle measures; do not stop without medical advice.
Headache, nausea, diarrhoea, abdominal pain, constipation, muscle pain (myalgia), weakness, increased liver enzymes
Contraindicated in active liver disease or unexplained persistent elevation of liver enzymes. Risk of myopathy/rhabdomyolysis—seek care for unexplained muscle pain, tenderness, weakness, especially with fever or dark urine. Use caution in liver impairment, heavy alcohol use, hypothyroidism, renal impairment, and in elderly patients. Liver function and lipid profile monitoring may be advised.
Strong CYP3A4 inhibitors (clarithromycin, erythromycin, ketoconazole/itraconazole, HIV protease inhibitors), cyclosporine, gemfibrozil/other fibrates, niacin, colchicine (increased myopathy risk), warfarin (may alter INR), digoxin (levels may increase), grapefruit juice (increases atorvastatin levels)
